Growth, Sporulation, Conidial Germination and Lethal Temperature of Paraphoma radicina, A Fungal Pathogen of Alfalfa (Medicago sativa) Root Rot

In 2020, alfalfa root rot, a disease caused by Paraphoma radicina, was identified in Inner Mongolia, China, where it seriously affected crop yields. Conditions for vitro growth, sporulation and conidial germination of P. radicina are poorly understood, limiting further studies. this contribution, we evaluated the suitability different media, carbon nitrogen sources, as well temperature pH growth germination. addition, sensitivity these cultures assessed. were most vigorous on ARDA medium, reaching maximum rates after 4 weeks incubation. All sources supported but none induced sporulation. The best mannitol peptone, respectively. Conidial observed to 35 °C range, with an optimum 25 °C. rate highest at 7, more than 50% conidia germinated 38 h incubation On other hand, temperatures above 55 (10 min) 41 proved lethal mycelial forms pathogen, These results can provide clues environmental conditions amenable infection crops and, whole, better understanding pathogenicity.
